I wouldn't do it. (Not that I am a big expert, not yet )
What if Google suddenly decides that you are cheating and kicks you out of the programm? (even if you are innocent, try telling this to them) Also if there are not that many adwords advertisers in your niche, it might happen that most of them decide not to go with content advertisement. It is recommended in books on Adword and I assume more and more people will read the books.
Saying that, I would design a site that is primary focused on Adsence but can easly be repurposed if something goes wrong with Adsence. _________________ Good luck to you all!

Tanya's last sentence is the way that I go about building my sites.
I have lots of content, links to affiliate programs, and my Adsense ads featured prominently. My sites make most of their money with Adsense.
However, if Adsense went away tomorrow, what am I left with?
An original-style Martell site, with lots of good content, and links to affiliate programs!
Design with Adsense in mind, but make sure your sites are flexible enough to earn money in another manner without having to make major changes to the sites.
Be sure to experiment also.
Adsense does take away potential visitors who might buy through your affiliate link. If you remove Adsense, your affiliate sales will almost surely go up, but will they go up enough to surpass your Adsense income?
In some niches, absolutely. In other niches, not even close.
The only way to know for sure is to test and experiment.
